The monarch shown here is a wink at "Mad" George III (who likely suffered from bipolar disorder).
Bugs has a nephew here, Clyde. In cartoons and films of the time, the censors demanded marriage of a character before there could be children. Having a niece or nephew appear, however, was a clever way to show the main character's parenting skills without requiring matrimony. Popeye and Olive Oyl, Mickey Mouse, and Barney Bear also had such occasional sidekicks.
